Effect of Uncaria Rhynchophylla on Expression of pERK1/2 and cFos Proteins in the Vestibular Nuclei Following Acute Arterial Hypotension

Abstract

|
The purpose of this study was to evaluate effect of Uncaria rhynchophylla, a Chinese herb, on expression of pERK1/2 and cFos proteins induced by acute hypotension in the medial vestibular nucleus (MVN) of Sprague-Dawley rats. Acute hemorrhage with 4 ml loss of blood resulted in significant synthesis of cFos-like protein in the MVN in which the number of cFos-like immunoreactive (cFLI) neurons was much higher in the caudal MVN in rostral aspect. U. rhynchophylla treatment of 500, 1000 mg/kg resulted in significant reduction of cFLI neuons following acute hypotension in the MVN. Single injection of U. rhynchophylla with 250 mg/kg caused significant reduction of pERK immunoreactive neurons activated by acute hypotension in the MVN. After U. rhynchophylla treatment of 500, 1000 mg/kg injection there was also inhibited markedly expression of pERK immunoreactive neurons in the vestibilar nuclei in response hemorrhage compared with saline treated animal. These results suggest that U. rhynchophylla inhibit increased excitability of neuron in the vestibular nuclei receiving normally excitatory afferent inputs from vestibular end- organs (Supported by a grant of the Korea Health 21 R&D Project, Ministry of Health & Welfare (02-PJ1-PG3-21302-0010).
